Original abstract
<h4>Introduction</h4>Cognitive impairment associated with neurodegenerative conditions is frequently driven by oxidative stress and neuroinflammation arising from metabolic and environmental insults. Streptozotocin (STZ) and aluminum chloride (AlCl₃) are widely used to induce experimental models of cognitive dysfunction through distinct pathogenic mechanisms. This study evaluated the neuroprotective effects of the probiotic <i>Lactobacillus acidophilus</i> in these two models.<h4>Materials and methods</h4>Adult male Wistar rats were divided into five groups: control, STZ, AlCl₃, probiotic + STZ, and probiotic + AlCl₃. Rats received intracerebroventricular STZ or saline, AlCl₃ in drinking water, and daily oral gavage of <i>Lactobacillus acidophilus</i> for 14 days. Behavioral performance was assessed using the novel object recognition and passive avoidance tests. Aluminum levels, oxidative stress markers, inflammatory cytokines, and hippocampal neuronal integrity were evaluated using biochemical and histological analyses.<h4>Results</h4>STZ and AlCl₃ administration induced significant memory impairment, increased oxidative stress and pro-inflammatory cytokines, and reduced the number of healthy hippocampal neurons. Probiotic treatment significantly improved memory performance, reduced lipid peroxidation and inflammatory markers, enhanced antioxidant defenses, decreased aluminum accumulation, and preserved neuronal integrity in the hippocampus.<h4>Conclusion</h4>Oral administration of <i>Lactobacillus acidophilus</i> effectively attenuates cognitive impairment, neuroinflammation, and oxidative stress induced by both metabolic and metal-related neurotoxic insults. These findings suggest that probiotic supplementation may represent a promising preventive strategy against cognitive dysfunction associated with diverse neurotoxic conditions.
